The Depo is not implemented yet. You only need it lvl 1 to activate ACS Defense , it doesn't take any deut though.

I see everyone doing the same thing and I would like to take a differen approach. One I have hyperspace engins will I be able to send a colony ship to another Galaxy?

No, but you can send them anyway, and if it's too far for it to go alone then send some LC with it.

you dont need hyperspace engine for colony ships, just go to the technology tree adn it will show you what you need to create a colony ship. Once built, it only takes the correct amount of deuterium to send it away. Longer distances may require the colony ship to be accompanied by a cargo ship as weresloth said.

I would like to know if when you colonize you add the required resources (What are they if so?) in addition to the Det to make the trip?

Never send the res with the colony ship, always send it after. If you send it with the colony ship, you lose it.
